Using sterile water instead of bacteriostatic water for multi-dose use

Sterile water has no preservative. It's safe for a single draw from a vial used immediately. Used for a multi-dose vial across 30 days, each needle insertion introduces contamination risk that compounds without the benzyl alcohol to suppress bacterial growth. The reconstituted solution looks identical to a correctly prepared vial. Use bacteriostatic water unless you're using the entire vial in one sitting.

Adding BAC water directly onto the powder

Directing the water stream onto freeze-dried peptide creates turbulence and pressure at the powder surface. For sensitive peptides, this can cause mechanical disruption of the peptide structure. The correct technique: angle the vial so the needle tip touches the glass wall and release the water slowly so it runs down the side to the powder. The peptide dissolves as the liquid level rises around it, not under high-velocity impact.

Shaking the vial to dissolve the peptide

Shaking generates foam and introduces mechanical stress. Protein-based peptides can aggregate or partially denature under vigorous agitation. The correct motion is a slow circular swirl — gentle rotation until the powder visually dissolves. Most peptides dissolve in 30–90 seconds with swirling. If the solution is still cloudy after 3 minutes of gentle swirling, the peptide may be degraded or the BAC water compromised.

Using the wrong BAC water volume and not recalculating

If you add 4mL instead of 2mL, your concentration drops from 2,500 mcg/mL to 1,250 mcg/mL. Your previously calculated 10-unit draw now delivers half the intended dose. The solution is visually identical. The only way to catch this is to recalculate before every new vial. Use the reconstitution calculator each time — don't rely on memory from a previous vial.

Freezing a reconstituted vial

Lyophilized (unreconstituted) peptide can be frozen safely. Reconstituted peptide in BAC water should not be frozen — ice crystal formation can disrupt peptide structure and benzyl alcohol precipitation at freezing temperatures alters the solution's preservative chemistry. Store reconstituted vials at 2–8°C (refrigerator) for up to 30 days. Freeze only unreconstituted dry powder for long-term storage.

Reconstitution Problem Questions

How do I know if my peptide is degraded?
Visible cloudiness or particles in the solution after proper reconstitution can indicate degradation. A correctly reconstituted peptide in BAC water is clear and colorless — similar in appearance to water. However, degradation is often not visually detectable. If you suspect degradation (peptide didn't dissolve normally, was stored improperly, exceeded 30 days after reconstitution), discard the vial. Degraded peptide is not dangerous but produces reduced or unpredictable effects.
Can I re-reconstitute if I used the wrong BAC water amount?
No. Once you've reconstituted, you cannot reverse the process or change the concentration by adding more BAC water — you'd need to know exactly how much was already added to maintain accurate dosing, which is impractical. If you added the wrong volume, document the actual BAC water amount used and recalculate your draw units based on the actual concentration. Do not add more BAC water to a reconstituted vial.
Why did my peptide turn yellow after reconstitution?
Slight yellowing is normal for some peptides — particularly those containing cysteine or methionine residues, or those stored for longer periods. A faint pale yellow is generally acceptable. A deep yellow or orange color, or any color change that was not present at initial reconstitution, suggests possible degradation or contamination and warrants discarding the vial. When in doubt, discard.
